Synthetic biology

Gene-editing therapeutics now deliver – Casgevy is a real product – but the synbio platform-foundry model still burns cash, so the value sits in tools, drugs and DNA supply, not the foundries.

The thesis

Core thesis

Jason Kelly (Ginkgo Bioworks) built the 'AWS of synthetic biology' — a platform for designing custom organisms. Emily Leproust (Twist Bioscience) revolutionised DNA synthesis cost. Applications span pharma, agriculture, food, materials, and industrial chemicals. But commercial revenue growth has been slower than expected — Ginkgo's stock fell 90%+ from IPO. The technology works; the go-to-market is the challenge.

State of the art (2026)

By mid-2026 synthetic biology has split into a working therapeutic wing and a still-unproven platform wing. Casgevy (Vertex/CRISPR Therapeutics) has moved past 500 patients initiated globally, ~90% reimbursed in the US, and roughly $43m in Q1 2026 sales – the first CRISPR drug to behave like a real product. In-vivo editing is harder: Intellia paused both nex-z Phase 3 trials after a patient death and an FDA hold in late 2025, with both holds since lifted and a BLA targeted for 2028. AI-for-biology is compounding fast – ESM3 in Science, David Bakers 2024 Nobel, RFdiffusion3 open-sourced. The platform-foundry model still bleeds cash: Ginkgos 2025 revenue fell 25% to $170m and burn continues.
